Yes. 256kbps mp3 and 320kbps are already a world of difference. The difference between 320kbps and FLAC is simply unnoticeable if encoded properly.

Nope. I personally stick to FLAC to have master copies so I can convert them to MP3 v0 for my portable player

Actually no. Depends on what kinda music you listen to and the master process.
If a song was mastered like shit, then the compression from the MP3 will not sound much different from the loseless compression from the FLAC.
Remember that mostly the advantage of FLAC is that it compresses masters without compromising the bitrate, so if the master sucks the compression method won't make much of a difference.
Still, if you ever could, I'd advice you to listen to Daft Punk's Random Access Memories with good headphones in FLAC format against the MP3 256kbps with whatever. There really is a world of difference.
