Nope. The Dome of the Rock is over the remains of the Roman temple that was inside Fort Antonia. What is currently officially called Fort Antonia by the Israeli government couldn't have possible held a 10,000 strong garrison. The entire "temple mount" precinct was Fort Antonia.
The Second Temple was about 200m south of the Al-asqa mosque, over the Gihon Spring in the City of David.
