Depends on how Iowa and New Hampshire go. It's unlikely Trump loses the nomination, but if a challenger does well enough it could damage Trump in the general election. Look at the last 4 Presidents who lost after their first or first and a half term: LBJ, Ford, Carter, and Bush. What do they all have in common?

Strong challengers. LBJ declined to run again in 1968 because of a poor performance in New Hampshire. In 1976, Reagan very nearly beat Ford in the GOP primaries. Ford lost. In 1980, Kennedy did VERY well against Carter and winning many states. Carter lost big league to Reagan. In 1992, Buchanan didn't win any states but still got 20%+ of the vote and damaged Bush heading to the general.

So a challenger doesn't necessarily need to beat Trump, but if one emerges they could damage Trump heading to the general. Remember, Trump didn't win the majority of votes in the 2016 primaries and lost the popular vote in 2016 general. Trump is pretty unpopular as well - if the GOP loses the House then I can see a challenger emerging in 2020.
