It's more to mock the drive for purity among hypocritical Americans who are absurd mixes of Europeans and who often have a small percentage of any possible race, with all the weird culture over "exoticism" via great-great-great whatevers.
A good part of the meme is your low standards of culture, a good part of it is the racial confusion, but the meme as a whole is directed at mocking the schizophrenia and lack of self awareness that characterises all aspects of America, racial and cultural. It's more open to context and interpretation than most memes in how it's used.
